The mother, child and assumed father should all be tested. If a sample cannot be obtained from the mother the testing can still be performed, however the results will be less conclusive than when both parents are tested.

To make the most accurate assessment, it is desirable to have samples from all parties potentially involved in the biological relationship. In the case of paternity testing, this would require a sample from the mother, alleged father and child. However the test can be performed with just the samples from the alleged father and child. For more complicated cases, assistance can be sought from the laboratory as to whom should be tested.
