The earliest versions supported are Windows 2000 Professional and Office XP, so it is definitely worth upgrading your desktops to WSUS-supported configurations. The optimal configuration would be desktop PCs running Windows XP SP2 and Office 2003. In addition, depending on what features you want to use with SBS 2003 R2, you will want to upgrade to the latest versions. If you want to take advantage of team workspaces, version control, or other SharePoint features, you must use Office 2003 or later.
