Why is it that, when the Mastercycler display shows the target temperature has been reached (e.g. from 72°C to 94°C), the cycler does not immediately begin counting down the time for that step?
The Mastercycler displays the temperature of the block on the lower left-hand corner of the screen as programmed T°C/ actual T°C. If a time lag exists, then the program is running under Tube Control. Tube Control is a preprogrammed method that ensures the sample within the PCR tube is heated to the set temperature. Since the actual sample temperature is attained later than the block temperature, countdown begins when the sample temperature has been reached in the tube.
