Blank is not always prepared with distilled water,it is prepared basing on the solvent of stock solution, that is Eg:In glusoce estimation, for the stock solution preparation know amount of Glucose is dissolved in the water. For this experiment Blank is prepared with distilled water. Use of Blank reading: Blank reading gives only reagents concentration (without solute concentration). When we substract this blank reading from test readings, finally we get concentrating of solute.
