Most likely there is no cell growth in some areas because of poor mixing of the Gelin-S/Glycosil/Extralink. Glycosil only will not support cell attachment. Try vortexing the Extracel components before plating. This is especially important if you are making Glycosil-only hydrogels with non-covalently incorporated extracellular matrix proteins.
