You can do a quick format in just a few minutes- all it does is delete the old list of files and add the structure NTFS uses to index the system. It is not just for bigger drives only. The advantage of FAT32 is that it is understood by all operating systems and devices- Windows, Mac, Linux, XBox, PS3, and Wii. NTFS can only be read and written to by Windows. Mac and Linux can read it, but not write to it (unless you get a separate driver). The PS3 and Wii cannot understand it at all. The disadvantage of FAT32 is that it can only hold 4GB files at the most. Also, it will not work on drives larger than 2TB.
