When are the Mastercycler Wait and Nowait modes used for heating the lid?
For most PCR experiments, the Nowait setting should be selected. However, especially when very small sample volumes are used (< 10 µl), it is sometimes better to wait until the lid has reached the required temperature. When this is required, it is even better to preheat the lid under the menu item Lid, and then place the samples and run the PCR only after the required temperature is reached.
