What volumes are best for multiwall plates?
The working range for 384 well plates is about 10 uL – 100 uL, and 25-250 uL for 96 well plates. When working with live cells, be careful not to remove all of the liquid such that the cells become dry. Ensure that there is a sufficient residual volume of liquid covering the cells between each fluidic step by observing the wells – this is typically no less that 10-15 uL/well in 384 well plates and 25-30 uL/well in 96 well plates and depends on the viscosity of the liquid and dimensions of the well. Measure your ideal residual volume and adjust the starting concentration of your subsequent reagent addition to account for this dilution.
