What temperature is best for hybridization on Biodyne nylon membranes?
Hybridization temperature is determined by the amount of selectivity desired. Lower temperatures (or hybridization stringency) will allow hybridization of sequences with less than 100% homology. Increasing stringency (by increasing temperature, decreasing salt concentration or addition of formamide) will assure that only completely homologous sequences hybridize. Of course, once the melting point of the double stranded complex is exceeded (this will vary with the other stringency factors in the solution), no hybridization can take place.
