Centrifuge the BD CPT™ Tube with blood sample at room temperature (18-25°C) in a horizontal rotor (swing-out head) at 1,500 to 1,800 RCF for a minimum of 20 minutes. Note: Collection of cells immediately following centrifugation will yield best results. Alternatively, the separated mononuclear cells can be gently resuspended into the plasma for storage or transportation for up to 24 hours. After storage/transport, pipette the entire sample above the gel layer into a separate tube to perform cell-washing steps.
