The easiest and cheapest way to stain cells is to add a DNA stain directly to a formaldehyde fixation solution and NOT to remove the residual media from your cells prior to a fixation. Try adding Hoechst 33343 into a formaldehyde solution so that the final concentration on the cells is between 1-10 uM Hoechst 33343 and 2-4% formaldehyde. Let the solution equilibrate for 1 hour before imaging. There will be some residual background from the Hoechst in solution, but it is typically negligible.
