Difference is just in the method of sample collection. Any method of sample collection do not affect the results of DNA paternity testing in any way. A buccal swab sample is just as accurate as a blood sample. The advantages of using buccal swab collections are that they are non-invasive, painless, quick and easy. Moreover, RBCs in humans do not have nucleus. So we can not isolate DNA from RBCs. As a result we need large amounts of Blood to collect DNA containing WBCs and platelets which are relatively lesser in number.
