What is the advantage of using aliquoted CyDye provided with the CyScribe Post-Labeling Kit instead of mono-reactive CyDye from the BioDirectory™?
The dyes available in the BioDirectory are formulated for use in relatively large-scale protein labeling; hence, each vial potentially contains dye for several cDNA labelings. The excess dye is then either thrown away, or needs to be dissolved in an organic solvent, freeze-dried and then stored aliquotted for future use. However, this renders the reactive dye susceptible to degradation by the air humidity—even if stored in closely capped tubes. Tests have shown that the content of reactive groups decreases by about 1% per day when aliquots are stored. This means that these aliquots have a very limited shelf life. The CyDye aliquots in the CyScribe Post- Labeling Kit are sealed in foil packs with desiccant. Finally, the reactive CyDye supplied with this kit are tested for suitability for the cDNA application whereas the dyes listed in the BioDirectory are only tested for protein labeling.
