The advantage is that you can perform live cell experiments (to look for dynamic processes such as protein translocation, drug efflux rates, changes in membrane potential etc) and cross- correlate the data with post fixation immuno-staining (to identify sub populations of interest i.e. distinguish between CD4+ and CD8+ lymphocytes etc.).
