What can be done, if the homogenate in buffer W1 cannot be transferred using a pipette?
The water content in leaves highly differs. If the leave material is always homogenised 1:1.5 in buffer W1 this sometimes may yield a very viscous mass, which cannot be pipetted even with wide bore tips. In this case the proportion of leaf weight to buffer has to be changed to 1:3. This causes a better extraction of DNA and yields nearly the same amount of DNA.
