Adenoviruses after CsCl purification are stored in a buffer containing 10 mM Tris-HCl, pH 8.0, 2 mM MgCl2 and 4% sucrose. Viruses in this buffer should be stored at -80°C and repeated freeze-thaw cycles should be avoided because viral titer will significantly decrease each freeze-thaw cycle. Viruses purified by chromatography are stored in a buffer containing 20 mM Tris/HCl, pH8.0, 25 mM NaCl, and 2.5% Glycerol (w/v). Viruses are more stable in the presence of glycerol and virus titer is not affected by several freeze-thaw cycles. Viral crude cell lysate (e.g. viruses in DMEM supplemented with serum) can be stored at -80°C and it will remain stable for several years and through several freeze-thaw cycles.
