What are the DNA quality recommendations for use with the DualChip GMO Kit?
DNA quality suitable for a reproducible qPCR experiment can generally be used with the DualChip GMO Kit. The quantification of the extracted DNA can be performed using the “Quant-iTâ„¢ PicoGreen dsDNA Assay Kit” as recommended by the supplier (Invitrogen). Dependent upon the matrix used, different DNA measurement methods might lead to different results. To control for contaminants that might inhibit the PCR reaction, photometric readings at 230 nm, 260 nm and 280 nm are recommended. UV-absorbing contaminants such as sugars, salts and organic solvents (absorb at 230 nm) and proteins (absorb at 260 nm) can be estimated. Pure DNA has an A260/A230 ratio of 2.5, and an A260/A280 ratio of 1.7 to 2.0.
