What are the advantages and disadvantages of Reverse Phase Chromatography?
The major advantage of RPC is resolution. Although the exact mechanism is still not well understood, the selectivity of the technique is excellent. Manipulation of parameters such as pH, ion-pairing agent, nature of modifier, etc. results in high-resolution separation of small and large biological molecules. Disadvantages of the technique are low capacity, denaturing conditions, and obligatory use of organic reagents such as TFA and acetonitrile, which are hazardous and present difficult disposal issues.
