I think you can achieve that in Macromedia Director. Works quite well with Flash and Fireworks. It allows for vector animation (transform, skew, movement, blend transition etc) over an image or animated video. In addition to that, you can also save a ‘behaviour’ such as ‘mouse over’ on the object. BTW For best movie trailers, you can’t expect to get all done in Director, though. Director only have a very limited 2d / vector drawings capability. You will need at least Gimp / Photoshop to make your image looks good (or Maya / Max / Blender if you need a 3d effect), and then rely on Director for the effects only. For web, what you need is to optimize your files. No use in creating stunning animations that takes long time to load. Check out the commercial websites and take note of their filesize / loading time.
