(a) Miniplasmid DNA must be RNase and Protease A treated before phenolextraction and ethanol precipitation. (b) Analytical preparations of DNA are preferable, such as CsCl gradient or one of the commercially available kits: Qiagen, Promega Maxi Kit, etc.
