Is the Harbor Main St site likely to incur more site and construction costs than other site?

No. In relation to the other sites considered, this site will require disturbance of less area with less soil materials to move/remove due to its topography. There will be no off-site sidewalks to build, no long entry driveways to build, very short water and sewer extensions from street to build and less parking to build. The current LSPA / Harbor Riverway office building will need to be removed and perhaps moved and reused elsewhere. The proposed building is conceptually similar to the previous Ski Tow site building, so building costs should be similar, but with inflation cost increases. Both are 2 story with the first floor built into a slope.
