Yes. Some samples such as corn starch contain low amounts of DNA and require larger sample sizes for testing (2 g). In this case a hydration test is appropriate before beginning. When using such sample sizes it is ideal to recover 600-800 µL of supernatant after centrifugation to go into Step 6.
