This is used in flies. C. elegans has a different mechanism: start with an m/m; ncl/ncl animal carrying a “free duplication (Dp)” of m+ ncl+. The free Dp is not attached to a chromosome and lacks a centromere, so it will be spontaneously lost during mitosis and all daughter cells will lack the gene. ncl is the cell autonomous marker, which gives large nucleoli.
