No, WordPress is built in PHP language but to use it knowledge of PHP is not must. The only time knowing PHP would be helpful while installing a plug-in and changes in some of the files are needed. This would not be the case normally as most of the plug-in comes ready to use and with a readme file. Other than installing plug-in, changing in the out of the box code would not be required.
