The only differences between these vectors is the presence of the coding sequence of the signalpeptide in pMM1525, pHIS1525 and pSTREP1525. That means it is possible to clone the gene of interest in the MCS in parallel (in pHIS1525 downstream of the coding sequence of the signal peptide). Please clone into the right reading frame as marked by the Start codon for xylA.
