In what formats will shRNA be available for pickup from the UMCCC shRNA Core Facility?
Both the human and mouse shRNA libraries are available from the shRNA Core Facility in any of five forms: Individual clones, individual purified plasmid, 960-plasmid general pools (960 plasmids per pool, random selection), user-designed custom pools, and as lentivirus. Individual clones will be provided in 5ml aliquots with 100ug/ml Carbenicillin and 25ug/ml Zeocin. Growth should always be performed with both of the above antibiotics, at stated concentrations, to ensure accurate and effective bacterial selection. Upon delivery, we recommend storing 1ml of this aliquot as an 8% glycerol stock, at minus 80C, which should always be streaked for fresh single-colony selection. Individual plasmids will be provided in approximately 100-300ug transfection grade aliquots, as required by the Vector Core for lentiviral production. 960-plasmid pools will be provided in 50ug transfection grade aliquots, sufficient for 5-10 genetic screens.
