If the PSEngine is stopped on the Plotstream Plot Server, can I still send agents to the queue?

Yes. The agents will stay in the queue until the PSEngine is started. Even if the server is rebooted, the agents will remain in the queue until they are processed or deleted using the PSQueue component.
