How was the Hannon shRNA library constructed?
In early 2004, Paddison et al described their strategy and vector components used to prepare their shRNA libraries in the pSHAG-MAGIC1 vector (Short Hairpin Activated Gene silencing – Mating Assisted Genetically Integrated Cloning System, version 1). Further understanding of RNAi mechanisms and how to exploit RNAi pathways for enforcing silencing in mammals led to version 2 of the library in pSHAG-MAGIC2c (pSM2c; Silva et al.).
