The completed oligomers delivered by the Core Facility come to the user as a dry pellet. If you ordered purification by reverse phase (RP) cartridge, your oligomer is 85-90% pure with an ammonium cation as a counter ion. Shorter oligomers are usually come purer then longer oligomers. We recommend that purification by RP cartridge be carried out on all oligonucleotides shorter than 50 bases in length. This technique is a very cost-effective technique if you do not require HPLC purity. If you ordered purification by HPLC or preparative PAGE, your oligomer should be 90 to 95% pure. After the oligo has been purified by HPLC or PAGE, it will be desalted on a RP cartridge. If you order more then 10 ODU (300 ug) of 95% pure oligomer we purify it by Ion-Paired HPLC (Reference). After purification the product will be precipitated from 70% IPA in the presence of sodium acetate. RNA and DNA/RNA chimeras are always purified by Ion-Paired Chromatography.
