How often do I subculture the reference strains from the stock culture kept at ‑70C and how frequently should QC testing be performed using these strains?
A. At a minimum, QC should be performed at least once weekly (rarely used discs should be QC-ed in conjunction with susceptibility testing) or when any component of the test is changed (new batch of media or discs etc). Ideally QC is performed with each batch of susceptibility testing. QC testing requires a fresh subculture of the reference strain(s). To minimise genetic changes to the reference organisms we recommend that the subculture to be used for QC should be no more than 5 passages removed from the cryogenically stored culture (6 passages from the received ACM strain). We recommend subculturing the reference strain from cryogenic storage once monthly. The recovered strain is then repassaged weekly until the end of the month. After overnight incubation, the repassaged strains should be held at 4C for the remainder of the week. A fresh subcultured is made from those held at 4C the day before QC testing is to be performed.
