How long is the Cell Sedimentation Manifold in place on the slide?

We have noted considerable variability in the rates of attachment by different cells to different substrates. Some cell lines or primary cell cultures adhere readily to some substrates within 30 minutes; other cell lines (even of the same histopathology) may require up to 14 to 20 hours to develop a deposited monolayer beneath the channel in the Cell Sedimentation Manifold. Using an inverted microscope, the attachment or spreading status of the sedimented cells can be examined while the CSM is still in place. Removing the CSM prior to establishment of an attached, confluent monolayer as a delineated disk of cells beneath the channel frustrates migration measurements. After a few uses of the CSM with any cell line, very consistent, reproducible migration studies can be easily conducted.
