How is the well-stirred models hepatic clearance calculated?
A. We calculate hepatic clearance according to the well-stirred model (see “Clearance concepts in pharmacokinetics” – Rowland M, Benet L Z and Graham G G., J. Pharmacokinet Biopharm, pp123-236, 1973) using the following equation: where Qb is the hepatic blood flow, bpr the blood:plasma ratio, fuplasma the fraction unbound in plasma, and Clint instrinsic clearance.
