How does the Xi-Clone PCR Cloning System work?
The Xi-Clone system uses homologous recombination to clone genes of interest into linearized Xi-Clone expression vectors. Initially, the gene is PCR amplified using gene-specific primers containing unique Xi-Clone end sequences. Next, the PCR product is mixed with a linearized Xi-Clone vector, and the mixture is added to the SmartCells competent E.coli. After transformation, endogenous bacterial recombinases incorporate the gene of interest into the Xi-Clone vector via homologous recombination.
