How do you program the gradient function on the Mastercycler®?
The gradient can be programmed into any step of your PCR protocol. Once you have your program displayed on the LCD, move the cursor down to the step number in which you would like to have a gradient. Press the [Options] key. A list of several programmable functions will be displayed (e.g. time extension, ramp rate). The last function will be the gradient. You may need to use the arrow key to scroll down to view this function because it may not all fit on the screen at once. To program a gradient, at the G =°C, type the number of degrees that you would like the gradient to span on either side of the temperature programmed for that step. For example, if you have 55°C programmed for that step and you type in a gradient of 5°C, then the temperatures across the block will range from 50°C to 60°C. Exit the program, and the Mastercycler will prompt you to save the newly edited program at that time.
