Attach autoclavable hose (Teflon, Silastic or pure gum rubber) to the outlet connector, using a clamp. Outlet hose length should not exceed 4 inches. If inlet hose is to autoclaved at the same time, it must not be longer than the outlet hose. If the filtrate receiving vessel is no larger than 2 liters capacity and has a vent, it can be attached to the outlet hose and autoclaved with the holder. Otherwise, a connector that can be aseptically attached to the receiving vessel must be installed on the end of the outlet tube and autoclaved with the holder. Wrap the exposed ends of hoses or connectors with Tyvek to allow passage of steam. Fasten the paper with autoclavable tape. Open all vent valves and included flow control valves, and autoclave at 121 degrees Celsius for at least 35 minutes and no more than 45 minutes. Time and temperature are important.
