How can I digest GenomiPhi products with restriction enzymes?
A. For efficient digestion of amplified products with restriction enzymes, samples should be heated up to 95 C and slowly cooled down. Approximately 80% of the amplification product should digest. Some smears might be seen, as well as the remainder of the undigested high molecular weight that will remain in your gel wells (see figure below). Restriction digest of amplified DNA. T.
