How are radioactive decay rates determined and how is the half-life measured?

Well, I know that the half-life is measured by how long it takes for the compound to stop effecting whatever has been exposed to it (by stop I mean slow down – the moment it ‘turns’ and starts being less effective, but still effective, is it’s half life). But I don’t know how radioactive decay rates are determined.
