Yes, it is possible to program the device that is mounted on PICDEM2 Plus demonstration board if the device is already programmed with boot loader code. By default, both sample devices (PIC16F877A and PIC18F4520) which you receive along with demonstration board are not programmed with boot loader. So you need to have ICSP based programmer for programming the boot-loader code.
