In the version I’ve got, Macbeth says this when he’s alone on stage as Lady Macbeth has gone to put the dagger back at the murder scene. It could be seen as Macbeth asking the audience for their judgement on his deed. There’s also a link to the old custom of judges washing their hands to rid themselves of the blame when they condemn someone (ie Pontius Pilate after he passes judgement on Jesus). Whatever justification M’beth and LMbeth managed to convince themselves of prior to the murder, I think M’beth realises there’s not justification at all, no amount of cleansing can take away what he’s done.
