Certainly paint can be measured wet (preferably with a non-contact instrument). However, for most paint the color will change significantly as the paint dries. Thus you will either have to learn how the colors change and compensate for that when you make wet measurements, or simply wait for samples to dry before measuring them. The best way to evaluate the color of finished paint is to create a sample as close as possible to the intended application and then measure it after the paint has dried.
