Yes, the One Step Checkout uses the same internal functions as the original Magento checkout and will display all activated payment methods and shipping methods. You can even set a default shipping and payment method in the administration. If you have different payment/shipping methods available in different countries, you can also set a default country in the backend, so it will display the proper methods immediately upon arriving in the checkout.
