Can I use any chemifluorescence substrate?
• Substrates tested successfully: o HRP system, QuantaRed Chemifluorescence Substrate, Pierce, Cat# 15159, http://www.piercenet.com/products/browse.cfm?fldID=2505DF51-5056-8A76-4E58-75B0587B4D88 o AP system, AttoPhos® AP Fluorescent Substrate System, Promega, CatS1000, http://www.promega.com/catalog/catalogproducts.aspx?categoryname=productleaf_1416&ckt=1 • Substrates known NOT to work with the Optimiser: o EX/EM wavelength near or lower than 400 nm, such as QuantaBlue from Piercenet • What about any restrictions on any other reagents. • We are currently validating all assay reagents and will keep updating the list. • So far, a known issue is the type of blocking buffer. The blocking buffer must be very efficient (owing to high binding efficiency of the Optimiser).
