Either approach is acceptable, but there are advantages to limiting the number of volunteers. By choosing a specific donor pool, you can limit the pool to include people with Moderate or Strong immune responses only. Therefore, you will be able to characterize the expected results for each donor. If you choose a random donor each time, you can expect to get results that cross all three zones, Low/Moderate/Strong. A portion of those will fail the 240 ng/mL ATP cutoff for the control sample; therefore, the assay run is deemed unacceptable.
