Only on operating systems that support GPT (GUID Partition Table). See the Microsoft GPT FAQ for a list operating systems that support GPT. When you first connect to a raw LUN that is larger than 2 TB with a GPT capable OS, the disk will show up in disk management as unknown/not initialized. The disk signature wizard should pop up. If it doesn’t, just right click on the disk and select initialize. The wizard will split the disk into a bunch of 2TB MBR partitions, which is probably not what you want. Right click on the disk and select Convert to GPT. The end result should look something like this: You can now create a new partition in the normal fashion and end up with a partition greater than two terabytes. Remember, this partition will only be accessable to hosts that support GPT.
