Are Cytoskeletons Involved in Establishment of Cell Polarity in Rat Salivary Glands?
The effects of colchicine (Col) and cytochalasin D (CD) on the generation and maintenance of cell polarity were examined in rat parotid and submandibular gland acinar cells. Secretory granules of these cells were depleted by the administration of d,l-isoproterenol (IPR) but reaccumulated during a 12 hr period. When Col was administered 1 hr prior to IPR, cell polarity of subsequently restituted acinar cells was markedly destructed. Without IPR, Col exerted little effect on polaritys. The CD treatment failed to induce polarity changes. These results suggest that an intact microtubular system is involved in the generation but not maintenance of cell polarity.
