You will need 1.0N NaOH and 1.0N HCl to adjust the pH of the 1X Assay Buffer to 7.4. Use of probenecid is not recommended. Control of “leaky” ion channels or pumps can require the use of appropriate blockers. Several of these blockers have been tested with the FLIPR Membrane Potential Assay Kit and found to be quite compatible. Blockers tested include: barium chloride as a blocker of voltage-gated potassium channels; suramin and pyridoxal-5-phosphate as blockers of P2X channels; TTX as a blocker of voltage-gated sodium channel.
